  • عنوان البحث : Molecular Detection of Burkholderia cepacia and Comparative Study Between Medical Plant and Antibiotics against Bacteria Isolates from Cystic Fibrosis

    Background: One of the major deaths causes in the patients with cystic fibrosis (CF) is the impaired lung function. Lately, Burkholderia cepacia was considered as a significant opportunistic pathogen in such patients. Since the late 1970s, due to its increased isolation from CF patients, its ability to spread infections in the community of CF patients, its role in disrupting lung function, and its inherent resistance to multiple antibiotics. Those various factors are making the Burkholderia cepacia isolation one of the significant tasks in cystic fibrosis health-care environment. Objective: To identify the method and the complete recA nucleotide sequence, and to compare the isolated bacteria from CF by medicinal plants and antibiotics. Material and Methods: Bacterial identification of CF patients was carried out by standard microbiological methods and identification was carried out using biochemical tests Vitek2 compact system, and PCR identification to confirm the identification and evaluate the antibacterial activity in Mentha leaves and lemon peel extracts against pathogenic bacteria. Results: Biochemical tests and gene expression confirmed that Burkholderia cepacia and Pseudomonas spp strains were isolated from 50 sputum samples. These results indicate that Mentha leaves extract is better than lemon peel extract, compares with antibiotics reveal the resistance of these bacteria.

    Abstract Objectives: Studying the cytogenetic effects of Mytomycin-C by using 2mg/kg by study of (mitotic index, chromosomal aberrations and micronucleus assays) on mouse bone marrow cells (in vivo) and Studying the cytogenetic effects of crude extract of Curcuma longa on mouse bone marrow cells and Studying the ability of crude extract of C. longa in reducing the genotoxic effects induced by Mytomycin-C by combination treatment on mouse bone marrow cells. Material and methodology: Administrative Arrangement: analytical study to period (1 / 5 / 2011 to 1 / 1 / 2012)conducted in Al-Nahrain research Centre for biotechnology, Baghdad, Iraq . Methodology: distributed of mice on equal group each group contain five animals, negative control and injected Phosphate Buffer Solution only and positive control injecte Mitomycin - C 2mg/kg and four groups injected by ethanolic crude extract by uses concentration (400, 200, 100 and 50 mg/kg) depend on LD50 of curcumin extract and study

Materials have evolved through the introduction and development of various production methods to improve the quality of materials, and one of these methods is the method of producing nanomaterial’s that gave a quantum leap in the properties of materials compared to ordinary materials because of their distinctive effect on (engineering, medical and technological applications). Among the most important applications of nanotechnology in cancer treatment, the effect of this technology has been observed to treat and reduce cancer. There are multiple ways to manufacture nanomaterials, but one of the most important methods is the Sol-Gel method, So we will address the fundamentals of this process, its most important applications because This method has proven its economic and efficient to manufacture various nanomaterials in different shapes and is considered simple compared to other methods. This review deals with methods of manufacturing nanoparticles in general and the treatment of cancer.

  • عنوان البحث : Effect of the Concentration Levels of Growth Hormone and Insulin-like Growth Factor I on the Polymorphisms of the Il12p40 Gene in Lung Cancer Patients

    The prevalence of lung cancer as one of the most common cancers with the highest mortality rate is one of the most important health problems in humans across the world. Molecular research can provide valuable information about genetic changes associated with the pathogenesis of the disease that may be used to improve prognosis and treatment. The current study aimed to examine the genotyping utility of the Il12p40 (IL-12B) gene (rs3212227, A>C) polymorphisms and detect its relationship with the concentration levels of HGH and IGF-1 for the non-small cell lung carcinoma (NSCLC). This study investigated 67 cases with NSCLC (60 males and 7 females) and 28 healthy individuals as controls. The serum level of HGH and IGF-1 was determined using an enzyme-linked immunosorbent assay. Genotyping of the IL-12B gene polymorphisms (rs3212227, A>C) was carried out by polymerase chain reaction-restriction fragment length polymorphism. The serum levels of the HGH and IGF-1 were estimated, and the results of the IL-12B genotyping showed an increased risk of NSCLC. The homozygous wild (AA) genotype of the IL-12 gene showed that the risk of NSCLC was higher than that of the heterozygous (AC) and homozygous genotypes (CC). Moreover, a significant elevation was found in the serum levels of the HGH in the NSCLC patients, compared to the control group. The result showed that the IL-12 gene polymorphism was implicated in the pathogenesis of the NSCLC and directed several metabolic changes.

    According to the World Health Organization, respiratory disorders, such as “influenza infection, acute tracheal bronchitis, TB, chronic obstructive pulmonary disease, lung cancer, and nasopharyngeal carcinoma”, have a major influence on human health. While environmental and socioeconomic variables might impact the pathogenesis of lung and respiratory tract diseases, it is nevertheless important to further investigate genetic and epigenetic reasons since a great many respiratory illnesses have a genetic or epigenetic basis. CRISPR is made up of interspaced, regularly-spaced palindromic repeated sequences and related proteins that carry out the CRISPR system's duties, which are found in prokaryotes' immune systems. This technology may be used to target, alter, and control genes, making it essential in respiratory research. Cas9 systems enable preclinical modelling of causative variables implicated in respiratory disorders, to generate fresh insights into its operations. CRISPR is also used to hunt for respiratory functions and pathology-associated genes. Which may lead to the discovery of new disease causes or therapeutic targets. The genetic and epigenetic mutations and the disease-associated mutations could be edited using CRISPR/Cas9. This kind of personalised medicine, which might be combined with stem cell reprogramming and transplantation are additional methods, that support embryonic stem cell expansion, might lead to the creation of novel respiratory illness treatment options. The new and developing area of investigation of CRISPR gene editing is one that requires further study the challenges of its specialty and the need for effective and safe delivery strategies. In respiratory health research and treatment, CRISPR systems represent an important step forward, and the discoveries made possible by this technology are likely to continue.

    — In this experimental study, we focused on methods of infection control through traditional disinfectant, in an attempt to contain the spread of the disease, which is increasing around the world. Previous studies focused on the different versions of the Coronavirus that appeared from 2003, and the results were similar in terms of prevalence and ways to resist infection on contaminated surfaces, so we assume that these conclusions apply to the new version of the virus because it belongs to the same family of Corona. The study conducted on different disinfection solutions have shown that antiseptics contain The Sodium Hypochlorite of concentration (0.1%) is a magic way for killing covid19 or Coronavirus, this concentration was done by mixing different quantity of bleach and water to reach the right concentration of (0.1%) of Sodium Hypochlorite; of a mix (100 ml) of bleach bottle of (1% ) with (900 ml) of water, mix (50ml) of bleach bottle of (2% ) with (950 ml) of water, mix (33 ml) of bleach bottle of (3% ) with (967 ml) of water, mix (25 ml) of bleach bottle of (4% ) with (975 ml) of water, and mix (20 ml) of bleach bottle of (5% ) with (980 ml) of water. The (concentration 0.1%) effective against coronaviruses. According to various studies, if infected contaminated surfaces and areas are cleared with appropriate concentrations of these disinfectants, they reduce the number of infectious coronaviruses particles to only a few within one minute. So it is very easy to eliminate them with disinfectants of any type affect this virus and reduce its focus on surfaces, but It is preferable to be in suitable concentrations, to eliminate the virus.
